Chromosome aberrations as a measure of mutagenesis: comparisons in vitro and in vivo and in somatic and germ cells.

چکیده
Structural alterations to chromosomes constitute a significant portion of the genetic damage produced by most mutagenic agents. The pioneering experiments of Muller (1), in the case of ionizing radiations, and of Auerbach (2), in the case of nitrogen mustards, showed that chromosome aberrations are, quantitatively, a significant class of mutational events for both physical and chemical mutagens. The ultimate goal in any mutagen screening program is to be able to test potential mutagens directly on human genetic material or to extrapolate to man from other test systems with confidence. At present the easiest way to attain that goal is to use chromosomal aberrations as an index of mutational damage. It is quite simple to obtain a large number of homogeneous human cells, treat them with the agent in question, analyze the chromosomes at the subsequent mitosis, and, if desired, compare the results to identical studies done on other mammalian species. The technique employed in this instance

Background: Sperm chromatin insufficiencies leading to low sperm count and quality, infertility and transmission of chromosomal microdeletion and aneuploidies to next generations can be due to exposure to environmental pollutions, chemicals and natural or manmade ionizing radiation. متن کاملEffects of 35 Hz, 2 mT Magnetic Field on Peripheral Blood Lymphocytes of Human In Vitro and Rat In Vivo
In this research we have studied the effects of extremely low frequency magnetic field on the peripheral blood lymphocytes chromosomes of human in vitro and rat in vivo. By this means, we used 60 blood samples of 10 men as well as 40 rats. Of 60 human blood samples ,40 samples (in two group of 20) were selected as test groups and placed into the 2 mT and 35 Hz magnetic field. The blood samples,...

Background: Nicotine as a toxic agent in cigarette has detrimental effects on reproduction. The aim of this study was to evaluate effects of of nicotine on germ and somatic cells in adult mouse testis. Materials and Methods: Male mice were divided into four groups. Group A or controls, groups of B, C and D were treated with nicotine intraperitoneally in doses of 0.1, 0.2 and 0.4 mg/100 g body w...

Background: To evaluate the effects of hyperthermia (HT) on the frequency of chromosomal aberrations induced by a low dose of neutron or γ-rays in human peripheral blood lymphocytes. Materials and Methods: Blood samples were exposed to HT (41.5°C for 30 and 60min, 43°C for 15 and 30min), 10 cGy neutron or γ-rays, HT + neutron/γ, and neutron/γ + HT. After standard cell culture, harvestin...

Background and Objective: Hybrid cells are made from fusion of two or more somatic cells. After formation chromosomes are located in one membrane . . So nucleic condition of each fused cell has changes and two genomes and chromosomes interact with each other. Locating the genes in new nucleic and cytoplasmic condition and great chromosomal rearrangement in these new formed cells can affect th...
